Vesicle capture by membrane?bound Munc13?1 requires self?assembly into discrete clusters

چکیده

Munc13-1 is a large banana-shaped soluble protein that involved in the regulation of synaptic vesicle docking and fusion. Recent studies suggest multiple copies form nano-assemblies active zones neurons. However, it not known whether such clustering correlated with multivalent binding to vesicles or specific plasma membrane domains at sites zone. The functional significance putative also unknown. Here, we report nano-clustering an inherent property indeed required for bilayers containing Munc13-1. Purified reconstituted onto supported lipid assembled into clusters from 2 ˜ 20 as revealed by combination quantitative TIRF microscopy step-wise photobleaching. Surprisingly, only minimum 6 were capable efficiently capturing retaining small unilamellar vesicles. C-terminal C2C domain clustering, but efficient capture. This capture largely due electrostatic hydrophobic interactions between membrane.

Insertion of nanoparticle clusters into vesicle bilayers.

A major contemporary concern in developing effective liposome-nanoparticle hybrids is the present inclusion size limitation of nanoparticles between vesicle bilayers, which is considered to be around 6.5 nm in diameter. In this article, we present experimental observations backed by theoretical considerations which show that greater structures can be incorporated within vesicle membranes by pro...

Synaptic vesicle capture by CaV2.2 calcium channels

The fusion of synaptic vesicles (SVs) at the presynaptic transmitter release face is gated by Ca(2) (+) influx from nearby voltage-gated calcium channels (CaVs). Functional studies favor a direct molecular "tethering" attachment and recent studies have proposed a direct link to the channel C-terminal. To test for direct CaV-SV attachment we developed an in vitro assay, termed SV pull-down (SV-P...

Selfassembly of metallic nanoparticle arrays by DNA scaffolding

We report the self-assembly of metallic nanoparticle arrays using DNA crystals as a programmable molecular scaffolding. Gold nanoparticles, 1.4 nm in diameter, are assembled in two-dimensional arrays with interparticle spacings of 4 and 64 nm. The nanoparticles form precisely integrated components, which are covalently bonded to the DNA scaffolding. These results show that heterologous chemical...

Electron capture by finite-size polarizable molecules and clusters.

The effects of finite molecular target size are investigated for partial-wave selected capture of electrons by isotropically polarizable molecules and clusters within a generalized Vogt-Wannier model. It is shown how expressions for partial-wave selected capture probabilities of zero-size targets from Dashevskaya et al. (Phys. Chem. Chem. Phys., 2009, 11, 9364) can be modified to account for fi...
